Polydatin and cardiomyopathy
Diabetic cardiomyopathy is the main cause of increased morbidity in diabetic patients, no effective treatment is available to date. Polydatin, a resveratrol glycoside isolated from Polygonum cuspidatum, demonstrates antioxidant and cardioprotective activities. However, the therapeutic effects of polydatin on diabetic cardiomyopathy and possible mechanisms remain unclear. This study aimed to investigate the cardioprotective effects and underlying mechanisms of polydatin on hyperglycaemia-induced myocardial damage.
In diabetic rats, polydatin administration significantly improved myocardial dysfunction and attenuated histological abnormalities, as evidenced by increases in left ventricular shortening fraction and ejection fraction, as well as reduced cardiac hypertrophy and interstitial fibrosis. In cultured H9c2 cells, polydatin pretreatment dose-dependently inhibited glucose-induced cardiomyocyte damage.

The results of this research show that the cardioprotective effect of polydatin against hyperglycaemia-induced myocardial damage is mediated by inhibition of NADPH oxidase and NF-κB activity. The results may provide new insight into the mechanisms of polydatin to be a potential treatment for diabetic cardiomyopathy.
